The pURE vector series (pURE1-4) is designed for generation of DNA templates for protein expression in cell-free coupledtranscription/translation reactions. These vectors contain the T7 promoterand a ribosome binding site (RBS) at an optimaldistance upstream from the ATGstart codon. They also feature a downstream T7 terminator. The ATG start codonof a cDNA encoding your protein-of-interestmust be clonedinto the NdeI, NcoI or SphI site of pURE1, pURE2 or pURE3, respectively. The pURE4 vector is for blunt endligation at an EcoRV site. These vectors are compatible for generating expression templates forPURESYSTEM®,PUREfrex®andThe RYTS (Remarkable Yield Translation System).The pURE vector series are especially designed for generation of template DNA for in vitro transcription/translation kit, PURESYSTEM. These vectors for transcription and translation are constructed from T7 promoter, and ribosome binding site (RBS) at a suitable distance upstream from the start codon ATG. Therefore, the 5'-end of gene of interest which includes the ATG start codon must be inserted into NdeI, NcoI or SphI site of pURE1, pURE2 or pURE3, respectively. The pURE4 is available for blunt 5’-end ligation with EcoRV site. It is also necessary to confirm the direction of the gene of interest if it is inserted into single restriction site.
